抗原负载的树突状细胞介导的抗自身膀胱癌作用研究
Specific anti-bladder cancer cell effect mediated by dendritic cells pulsed with autologous carcinoma antigen
【摘要】 目的:研究膀胱癌肿瘤细胞冻融抗原负载的树突状细胞(DC)对特异性抗自身膀胱癌作用研究。方法:将膀胱癌肿瘤病人外周血单个核细胞来源的DC在体外用冻融负载,再与膀胱癌患者的经细胞因子诱导的杀伤细胞(CIK)共同培养,应用乳酸脱氢酶释放法观察其对自身膀胱癌细胞杀伤活性,与未负载肿瘤抗原的CIK组进行比较。结果:DC经抗原负载后介导的CIK对自身膀胱癌细胞杀伤性活性明显增强。结论:用肿瘤抗原负载可进一步提高DC介导的CIK细胞对膀胱癌细胞的杀伤活性。
【Abstract】 Objective:To investigate the immunostimulatory effect of dendritic cells(DC)pulsed with bladder cancer antigen.Methods:Dendritic cells from bladder cancer patients were pulsed by autologous carcinoma cell lysates,and then cocultured with cytokine induced killer cells(CIK)from bladder cancer patients.The cytotoxic activity was measured using a lactate dehydrogenase release assay,and compared with CIK.Results:The cytotoxic activity of two groups against autologous carcinoma cells was(61.25±7.43)%,(28.75±4.18)%,respectively.There was a significant difference between the two groups.Conclusion:Coculture of CIK cells with DC pulsed with autologous carcinoma cell lysates can increase the cytotoxic activity of immunologic effector cells against bladder cancer cells.Dc pulsed with bladder cancer antigen may have an important effect on immunotherapy protocols for bladder patients.
