【摘要】 目的观察黄芩苷、川芎嗪对哮喘大鼠气道壁重构和基质金属蛋白酶-9(MMP-9)及抑制剂-1(TIMP-1)表达的影响。方法24只Wistar大鼠分为4组:正常对照组,哮喘模型组,黄芩苷组和川芎嗪组。用卵蛋白(OVA)致敏制备大鼠哮喘模型。HE染色测气道壁及平滑肌层厚度,免疫组化染色测定气道壁MMP-9、TIMP-1、Ⅳ型胶原含量。结果哮喘模型组大鼠气道壁及平滑肌增厚,MMP-9、TIMP-1表达增多,与正常对照组相比差异有统计学意义(P<0.05);黄芩苷组和川芎嗪组大鼠气道壁及平滑肌变薄,MMP-9、TIMP-1表达减少,与哮喘模型组相比差异有统计学意义(P<0.05)。结论黄芩苷、川芎嗪可以通过调节MMP-9/TIMP1比例,减少气道壁胶原沉积和平滑肌增生而降低气道壁厚度。更多还原

【Abstract】 Objective To explore the effect of two herb ingredients(baicalin,ligustrazine)on airway remodeling and the expression of matrix metalloproteinase-9(MMP-9)and tissue inhibitor of metalloproteinase-1(TIMP-1)in asthmatic rats. Methods Twenty-four Wistar rats were randomly divided into four groups:normal control group(n=6),asthmatic model group(n=6),baicalin group(n=6),ligustrazine group(n=6).The rats were sensitized with ovalbumin to establish the asthma model.Lung tissues were sliced and stained with HE.The airway wall and airway smooth muscle thickness were measured by image analysis system.The expression of MMP-9,TIMP-1 and collagen type Ⅳ in lung tissues was observed by immunohistochemistry. Results Compared with normal control group,the thickness of airway wall and airway smooth muscle in asthmatic model group significant thickened,and the expression of MMP-9 and TIMP-1 significantly increased(P<0.05).Compared with asthmatic model group,the thickness of airway wall and airway smooth muscle in baicalin group and ligustrazine group significantly thinned,and the expression of MMP-9 and TIMP-1 significantly decreased(P<0.05). Conclusion Both baicalin and ligustrazine could decrease the deposition of collagen type Ⅳ and reduce the airway smooth muscle thickness by influencing the balance between MMP-9 and TIMP-1.更多还原
