反相高效液相色谱法测定酶促转化法制备的S-腺苷甲硫氨酸

【摘要】 反相高效液相色谱法测定酶促转化法制备的S-腺苷甲硫氨酸,采用Luna C18柱(250 mm×4.6 mm,5μm),流动相为100%0.02 mol/L乙酸铵,用乙酸调pH值至4.0;流速为1.0 mL/min,用二极管阵列检测器检测,检测波长为254 nm,在200~800μg/mL范围内线性良好(r=0.9997),检出限为2.3 ng。

【Abstract】 RP-HPLC method was established for the determination of S-adenosyl-L-methionine.The analysis was performed with a Luna C-18 column(250 mm×4.6 mm,5 μm)and 100% 0.02 mol/L ammonium acetate(pH=4 adjusted with acetic acid) as mobile phase.The flow rate was 1.0 mL/min.A diode arry detector was used and the detection wave length was 254 nm.The calibratin curve was linear over a range from 200~800 μg/mL(r=0.999 7).The detection limit is 2.3 ng.
