【摘要】 建立了采用反相高效液相色谱同时测定药材蟾酥中华蟾酥毒基、酯蟾毒配基和蟾毒灵含量的方法。采用反相C18色谱柱,以乙腈-0.1mol/L醋酸缓冲液(pH3.2)(体积比为50∶50)为流动相,流速0.8mL/min,在299nm波长下检测。华蟾酥毒基、酯蟾毒配基和蟾毒灵的线性范围分别为0.25~0.875μg(r=0.9990)、0.25~0.875μg(r=0.9991)和0.15~0.525μg(r=0.9990);平均加样回收率分别为100.3%、100.0%和98.0%,结果表明所建方法简便可靠。对不同来源的10批样品中的3种蟾毒内酯含量的检测结果显示,不同来源或批次的蟾酥药材中3种蟾毒内酯总含量差异较大。更多还原

【Abstract】 A reversed-phase high performance liquid chromatographic method for the quantitative determination of three bufogenins in toad venom was established.An Alltech Alltima C18 column(250 mm×4.6 mm,5 μm)was used.The mobile phase was acetonitrile-0.1 mol/L acetate buffer(pH 3.2)(50∶50,v/v),and the flow rate was 0.8 mL/min.The detection wavelength was set at 299 nm.The method has good linearity in the ranges of 0.25-0.875 μg for cinobufagin(r=0.9990),0.25-0.875 μg for resibufogenin(r=0.9991),and 0.15-0.525 μg for bufalin(r=0.9990).The average recoveries were 100.3%,100.0% and 98.0% for cinobufagin,resibufogenin,and bufalin,respectively.The results indicate that the method is simple,accurate,reproducible,and can be used for the quality control of bufogenins in toad venom.更多还原
