芸豆蛋白质的提取及超滤分离研究

【摘要】 在单因素试验基础上用正交试验对芸豆蛋白的提取工艺条件进行筛选,得出最优提取条件为:在40℃下,用20倍于芸豆粉的pH10.00.067mol/L磷酸缓冲液,提取60min。同时探讨了芸豆蛋白超滤分离纯化工艺,结果表明,在压力差0.3MPa、泵功率20%、料液浓度4%、料液pH10.0条件下,经过四次超滤后,蛋白质纯度稳定在85%左右。

【Abstract】 Based on single factor tests,extraction conditions of protein from Phaseolus vulgaris were discussed and the optimum extraction conditions were determined by orthogonal test. The results of orthogonal test showed that the optimum extraction conditions are as follows:temperature 40 ℃,extraction time 60 minutes and proportion of materials to 0.067 mol/L PBS at pH 10.0 1:20. The application of ultra filtration technology in purification of protein was also discussed. On the conditions of pressure 0.3 MPa,pump power 20% and solution at pH 10.0,concentration 4% after four times ultra filtration,the protein purity is up to 85%.
