【摘要】 目的探讨热休克蛋白60(Hsp60)基因多态性和抗体水平及其与冠心病(CHD)的关系。方法采用病例对照研究,选取未治疗过的CHD患者615例,对照以年龄(±1)岁和性别以1∶1与CHD组匹配共选取615例。采用间接酶联免疫吸附法测定血浆Hsp60抗体水平;TaqM an-MGB探针检测hsp60基因上rs2340690、rs788016、rs2565163和rs2305560四个单核苷酸位点的多态性(SNPs)。结果CHD组的Hsp60抗体水平显著高于对照组,差异有统计学意义(P=0.000)。上述4个SNPs的基因型频率在CHD组和对照组之间差异均无统计学意义,采用多元Lo-gistic回归分析,对年龄、性别、吸烟、饮酒、高血压史、糖尿病史和家族CHD史等因素进行校正后,未发现4个SNPs的任一基因型与CHD的危险度有关联;多因素协方差分析校正了以上混杂因素后,CHD组和对照组hsp60基因4个SNPs不同基因型之间的血浆Hsp60抗体水平差异均无统计学意义(P>0.05)。结论Hsp60抗体可能与CHD发病有关,hsp60基因的多态性和抗体水平与CHD风险均没有显著性关系。更多还原

【Abstract】 Objective To investigate the heat shock protein 60(Hsp60)gene polymorphisms,and its relationship with the level of antibody to Hsp60(anti-Hsp60)and the risk of coronary heart disease(CHD)in Chinese.Methods 615 CHD without treatment and 615 age-and sex-frequency matched controls were selected for a case-control study.Levels of anti-Hsp60 antibody in plasma were measured by enzyme-linked immunosorbent assay,the genotypes of 4 tagged single nucleotide polymorphisms(tagSNPs)—rs2340690,rs788016,rs2565163 and rs2305560 were tested with TaqMan-MGB probes.Results The results showed that level of anti-Hsp60 was significantly higher in CHD group than that in controls(P=0.000),but there was no significant difference in the frequencies of genotypes of 4 selected SNPs between CHD group and control(P>0.05).After the adjustment for age,sex,smoking,drinking,hypertension,diabetes and CHD family history etc.with multivariant Logistic regression analysis,no any relation was found between plasma anti-Hsp60 level and genotype of the 4 SNPs(P>0.05).Conclusions The results suggested that the plasma level of anti-Hsp60 antibody might be associated with CHD,but there was no significant relation was found between polymorphisms in rs2340690,rs788016,rs2565163 and rs2305560 positions of hsp60 gene and the Hsp60 antibody level or the CHD risk in Chinese.更多还原
