红三叶寡肽酶解制备工艺研究
Study on enzymolysis technology for oligopeptide from trifolium pratense
【摘要】 本试验以水解度为指标研究不同蛋白酶对红三叶叶蛋白的水解作用。通过单酶筛选试验确定了中性蛋白酶Ⅱ(来源于1398枯草芽孢杆菌)为最佳水解酶,进一步的单因素试验和正交试验结果表明,酶解最佳工艺条件为酶解温度40℃,pH值7.6,酶活添加量5000U/g,底物浓度1%,酶解时间3h。
【Abstract】 Hydrolysis on red clover leaf protein with different protease was studied.Based on the screening experiments of each enzyme,neutral proteinase Ⅱ from Bacillus subtilis 1398 was selected as feasible enzyme for the decomposition of red clover leaf protein.The results of single factorial trials and orthogonal trial showed:the optimum hydrolysis conditions of red clover leaf protein were 40 ℃,pH 7.6,enzyme activity 5000 U/g protein,the substrates concentration 1 % and enzyme digestion 3 h.
