IL-1β对新西兰兔下丘脑前列腺素受体EP3亚型表达的影响

【摘要】 为了观察IL-1β对新西兰兔下丘脑前列腺素受体EP3亚型表达的影响,本实验用IL-1β复制新西兰兔发热模型,并用免疫组化方法检测兔下丘脑内侧视前区(medial preoptic area,MPO)和正中视前区(median preoptic area,MnPO)前列腺素受体EP3亚型的表达。结果表明:静脉注射IL-1β可引起新西兰兔体温升高,最大体温上升幅度(maximal raise altitude of temperature,△T)及1h体温反应指数(temperatu reresponse index,TRI1)均高于对照组;同时,注射IL-1β后,下MPO和MnPO前列腺素受体EP3亚型的表达明显增强。上述结果提示:MPO和MnPO神经元前列腺素受体EP3亚型表达的增强可能是IL-1β引起发热的中枢机制之一。

【Abstract】 To observe the effect of IL-1β on the expression of EP3 in the hypothalamus of rabbits. The expression of EP3 in the medial preoptic area (MPO) and median preoptic area (MnPO) of hypothalamus was detected by immunohistochemical method. The results showed that the colonic temperature raised gradually after i.v. IL-1β. The T(maximal raise altitude of temperature)and TRI1(temperature response index) of IL-1β treatment group were higher than that of control group. The expression of EP3 in the MPO and MnPO of IL-1β treatment group raised markedly compared with the control group. The present results demonstrate that the increase of EP3 expression in the MPO and MnPO after i.v. IL-1β may be one of the mechanisms of IL-1β-induced fever.
