HPLC和LC-MS~n分析葛根素及其注射剂中的有关物质

【摘要】 目的研究葛根素及其注射剂中的有关物质。方法采用HPLC对9批葛根素原料药和18批葛根素注射剂中的有关物质进行了系统考察,采用LC-MSn技术对葛根素及其注射剂中有关物质的结构进行了初步鉴定。结果葛根素及其注射剂中存在5种主要有关物质,其中2种为相对分子质量432的同分异构体,推测可能为3′-羟基葛根素和染料木素-8-C-葡萄糖苷。结论建立的方法可有效分离主成分葛根素与有关物质,对有关物质能进行初步鉴定。

【Abstract】 Purpose To study the related substances in puerarin.Methods HPLC-DAD method was used for detection of the related substances in 9 batches of puerarin and 18 batches of puerarin injection.LC-MSn method was used to characterize the structure of related substances.Results The study showed that there were five major related substances of puerarin,two of which were proposed to be genistein 8-C-glucoside,and 3′-hydroxypuerarin,and three others were to be identified through further study.Conclusion The method is simple and rapid.Good separation of puerarin from main related substances was achieved.And the possible structures of related substances were deduced.
