过表达番茄叶绿体小分子热激蛋白提高植株的耐热性
OVEREXPRESSION OF CHLOROPLAST-LOCALIZED SMALL MOLECULAR HEAT-SHOCK PROTEIN ENHANCES THE THERMOTOLERANCE IN PLANT
【摘要】 小分子热激蛋白是植物受到热胁迫后的主要表达产物之一,与植物细胞耐热有密切关系.我们将由CaMV35S启动子驱动的叶绿体小分子热激蛋白cDNA导入番茄,Northern及Western印迹分析表明,叶绿体小分子热激蛋白在转基因番茄中呈现组成型表达,通过测定转基因番茄和未转基因番茄的耐热性,发现转基因株系基础耐热性强于对照,说明叶绿体小分子热激蛋白的过量表达提高了植株的基础耐热性.
【Abstract】 The small heat shock protein is usually the most heat responsive of the HSP,and is tightly related to the thermotolerance in organisms.A DNA construction,including tomato chloroplast-localized small molecular heat shock protein cDNA under the control of cauliflower mosaic virus 35S promoter,was introduced into the genome of tomato plants.Both Northern-blotting and Western-blotting analyses indicated that the constitutive expression of Le-CPsHSP in transgenic plants.Upon high temperature,the transgenic plants exhibited stronger basal thermotolerance than that of control plant.These findings lead to the conclusion that CPsHSP can enhance the basal thermotolerance in plant.
