葛根素对血管内皮损伤后细胞外基质CatS、CysC表达的影响及意义

【摘要】 目的探讨葛根素对动脉球囊损伤后细胞外基质重塑过程中组织蛋白酶S(CatS)及其抑制剂胱蛋白C(CysC)表达的影响及其意义。方法建立兔髂腹主动脉球囊损伤后再狭窄模型。观察应用葛根素治疗前后再狭窄形成的范围。检测再狭窄局部CatS及其抑制剂CysC和内皮下胶原的表达水平。结果葛根素组与球囊损伤组比较,再狭窄范围明显减小,CatS表达水平降低,内皮下胶原含量减少,(P<0.05)CysC表达水平无显著差异(P>0.05)。结论葛根素可降低球囊损伤部位再狭窄形成过程中CatS表达、减少斑块内胶原的聚集,抑制再狭窄的形成。

【Abstract】 Objective To identify the significance and effect of Puerarin on Cathepsin S(CatS) and Cystatin C(CysC) in the procedure of vascular restenosis after vascular endothelial injury.Methods The experimental model of vascular restenosis in rabbits was established by balloon injury endothelium of Iliac abdominal aorta and then hyper-lipid feed.The arterious restenosis area,CatS and CysC,collagen differences were decfed.Results Compare with balloon injuried group,the restenosis area in puerarin group was obviously smaller(P<0.05);The express of Cathepsin S in puerarin group was smaller than that in balloon injuried group(P<0.05).The express of Cystatin C in puerarin group and balloon injuried group was not significantly different(P>0.05).Collagen in intima was less in puerarin group than that in balloon injuried group(P<0.01).Conclusion Puerarin can down-regulate the expression of CatS in the area of balloon injury during the procedure of restenosis and reduce collagen accumulation in the plaque to suppress restenosis.
