大脑皮层神经元微管相关蛋白-2在人胚发育过程中的表达变化
Fetal development phase variations of the microtubulin-associated protein expression in cerebral cortex
【摘要】 目的观察微管相关蛋白-2(MAP-2)在人胚胎大脑发育过程中的作用,为预防先天性神经系统疾病提供理论依据。方法收集3、4、5、6、7个月胎龄人胚胎组织标本,采用免疫组织化学方法观察其大脑皮层神经元MAP-2(a、b、c)表达变化。结果免疫反应阳性产物均定位于胞质,胞核不着色;随月龄增加,大脑皮层神经元MAP-2(a、b、c)表达逐渐增多,细胞阳性反应平均光密度(OD值)3月龄为0.1297±0.0051,4月龄为0.1832±0.0050,5月龄为0.2597±0.0058,6月龄为0.3284±0.0067,7月龄为0.3909±0.0039。结论在人胚胎大脑发育过程中,MAP-2(a、b、c)可能参与神经元的发育、结构稳定及突起形成。
【Abstract】 Objective To explore the fetal development phase variations of the microtubulin-associated protein expression in cerebral cortex.Methods Cerebral tissue samples of 3,4,5,6 and 7-month-old fetuses were collected.HE staining and MAP immunohistochemistry were used to observe the structure and MAP expression in cerebral neurons.Results The optical density(OD) of positive cellular reaction were as follows: with the 3-month-old,0.129 7±0.005 1;with the 4-month-old,0.183 2±0.005 0;with the 5-month-old,0.259 7±0.005 8;with the 6-month-old,0.328 4±0.006 7;with the 7-month-old,0.390 9±0.003 9.Conclusion Microtubulin may enhance the growth and structural stability of a neuron and regulating the formation and plasticity of the spines.
