泻火补肾汤对大鼠脑出血神经干细胞移植后脑内TGF-β1和VCAM-1蛋白表达的影响
Effects of Xiehuo Bushen decoction on expressions of TGF-β1 and VCAM1 in rats brain of neural stem cells-transplanted intracerebral hemorrhage
【摘要】 目的:研究泻火补肾汤对神经干细胞(NSCs)移植入脑出血大鼠脑室下区(SVZ)后转化生长因子(TGF-β1)和血管细胞间黏附分子(VCAM-1)表达的影响,探讨泻火补肾汤能否诱导移植免疫耐受。方法:将SD大鼠随机分为6组:A(正常组)5只,B(假手术组)、C(模型组)、D(模型治疗组)、E(移植组)、F(移植治疗组)各5只。采用免疫组织化学方法观察术后2 d4、d、7 d、14 d、28 d TGF-β1和VCAM-1的表达。结果:B、C、D、E、F五组VCAM-1和TGF-β1分别在2 d4、d表达达高峰(P<0.05)。VCAM-1在各时间点E组表达最强(P<0.05),而TGF-β1在F组表达最强(P<0.01)。同时间点除28 d外,F组比E组、D组比C组TGF-β1表达增强(P<0.05),而VCAM-1表达减弱(P<0.05)。结论:泻火补肾汤可通过促进免疫抑制因子TGF-β1的表达,抑制免疫相关因子VCAM-1的表达,从而有助于移植受体免疫耐受。
【Abstract】 Objective:To observe effects of Xiehuo Bushen decoction(XHBSD) on the expressions of transforming growth factor-beta1(TGF-β1) and vascular cell adhesion molecular-1(VCAM-1) in the rats brain of NSCs-transplanted intracerebral hemorrhage,and to investigate whether Xiehuo Bushen decoction can induce immunotolerance or not.Methods:One hundred and thirty Sprague-Dawley rats were randomly divided into six groups:A group(normal group)(n=5),B group(sham operation group)(n=25),C group(model group)(n=25),D group(treated model group)(n=25),E group(transplanted group)(n=25),F group(treated transplanted-group)(n=25).The expressions of TGF-β1 and VCAM-1 were assayed by immunochemistry 2 d,4 d,7 d,14 d,28 d after the operation.Results:The expressions of VCAM-1 and TGF-β1 in B,C,D,E,F groups arrived the peak at 2 d and 4 d respectively(P<0.05).The expression of VCAM-1 in E group is the highest among all the groups(P<0.05),while the expression of TGF-β1 in F group is the highest(P<0.01).Except 28th day,at the same time point the expressions of TGF-β1 in F and D groups were significantly higher than that in E and C groups respectively(P<0.05),and the expression of VCAM-1 in F and D groups were lower than E and C groups respectively(P<0.05).Conclusion:XHBSD can promote the expression of TGF-β1 and inhibit the expression of VCAM-1 in the rats brain of NSCs-transplanted intracerebral hemorrhage,and may induce the immunotolerance of the receptor.
