解毒化瘀方对脓毒症大鼠蛋白C mRNA表达及血浆蛋白C活性的影响
Effect of the Jiedu Huayu Recipe on the expression of protein C mRNA and the plasma activity of protein C in septic rats
【摘要】 目的观察解毒化瘀方对脓毒症大鼠凝血调节蛋白C mRNA(PC mRNA)表达及血浆蛋白C(PC)活性的影响,并探讨中药解毒化瘀方治疗脓毒症的作用机制。方法将大鼠分为正常组、对照组(假手术处理)、模型组(脓毒症)、中药组(造脓毒症模型前后用解毒化瘀方治疗),动态观察术后12、24、48 h及正常组肝脏组织PC mRNA表达及血浆PC活性、内毒素(LPS)水平。采用盲肠结扎穿刺法复制脓毒症模型、实时荧光定量PCR法检测PC mRNA表达、血凝仪法检测血浆PC活性,动态浊度鲎试验法检测LPS水平。且动态观察解毒化瘀方对脓毒症大鼠生存率的影响。结果在脓毒症形成过程中,模型组PC mRNA表达水平和血浆PC活性均显著低于正常组及对照组(P<0.01),而血浆LPS水平则显著高于正常组与对照组(P<0.01);经解毒化瘀方治疗的脓毒症大鼠,其PC mR-NA表达及血浆PC活性均显著高于模型组(P<0.01),血浆LPS水平则显著低于模型组(P<0.01);经解毒化瘀方治疗的脓毒症大鼠,其生存率亦高于未治疗的脓毒症大鼠(P<0.05)。结论解毒化瘀方能增强脓毒症大鼠PCmRNA表达,提高血浆PC活性,降低血浆LPS水平,提高了脓毒症大鼠的生存率。
【Abstract】 Objective To explore the changes of expression of coagulation regulating protein C mRNA(PC mRNA) and activity of plasma protein C(PC) in septic rats treated with Jidu Huayu Recipe(JHR).Methods Wistar rat sepsis models were induced by cecal ligation and puncture.The Wistar rats were divided into the normal group(NP),the control group(CP),the model group(MP) and the traditional Chinese medicine treated group(TCMP).The expression of Protein C mRNA and the activity of plasma PC and LPS were dynamically determined at 12 h,24 h,and 48 h after the sepsis operations by real-time quantitative RT-PCR and by the automated coagulation analyzer and Limulus test.The effect of JHR on the survival rate was also dynamically observed.Results In the course of sepsis, the expression of protein C mRNA and the activity of plasma PC in the MP group were lower than those in the CP and NP groups(P <0.01),but the LPS level was increased(P<0.01).The expression of protein CmRNA and the activity of plasma PC in the TCMP group were higher than that in the MP group(P <0.01),but the LPS level was decreased(P<0.0 1).The survival rate of the septic rats treated with JHR was obviously higher than that of those without JHR treatment(P<0.05).Conclusion JHR could improve the expression of protein C mRNA and the activity of plasma PC and reduce the plasma LPS level of septic rats,which improves the survival rate of septic rats.This may be one of the molecule bases of JHR in treating sepsis.
