阴离子交换色谱中色谱柱温度对氨基酸保留的影响

【摘要】 研究了阴离子交换色谱中色谱柱温度对氨基酸保留的影响。实验采用氢氧化钠和醋酸钠作为淋洗液在Dionex AminoPac PA10阴离子交换柱上分离氨基酸,色谱柱温度为20~40℃。氨基酸的保留表现为吸热行为(随温度升高,保留增强)或放热行为(随温度升高,保留减弱)。温度对弱保留氨基酸的保留有明显影响。利用温度对弱保留氨基酸保留影响上的差异,通过改变柱温可以改善两种相近洗脱氨基酸之间的分离状况。温度对强保留氨基酸的保留无明显影响。弱保留氨基酸的范特霍夫曲线(lnk与1/T关系曲线)具有良好的线性关系。

【Abstract】 Effect of column temperature on the retention of amino acids in anion-exchange chromatography was investigated. A Dionex AminoPac PA10 anion-exchange column was used to separate amino acids. Sodium hydroxide and sodium acetate were used as eluent. The column temperature was maintained at 20~40?C. Amino acids retention exhibited both exothermic and endothermic behavior. Significant selectivity variations of some weakly retained amino acids were achieved through changing column temperature. However, temperature was ineffective at changing the selectivity of strongly retained amino acids. The linearity of the Van’t Hoff plot for weakly retained amino acids is satisfactory.
