高效液相色谱法测定发酵乳中的维生素

【摘要】 研究了利用高效液相色谱(HPLC)法分离测定青海省传统发酵乳中维生素B6,B2和B1的条件。结果表明,流动相为0.005mol/L己烷磺酸钠—甲醇—乙酸(体积比为75︰25︰1),流速为1.0 mL/min,检测波长为280 nm,柱温为30℃时可以很好地分离测定发酵乳中的维生素B6,B2和B1质量浓度,该方法相对标准偏差小(0.45%~1.57%),回收率高(95.07~98.41%),线性相关系数高(r>0.999),具有较高的准确度和精确度。通过对青海省40份传统发酵乳(酸牦牛奶、酸山羊奶和酸马奶)中的维生素B6,B2和B1质量浓度测定发现,发酵乳样品之间维生素B6,B2和B1质量浓度差别较大。

【Abstract】 Studied on the condition of separating and detecting vitamin B6,B2 and B1 of fermented milk samples by high performance liquid chromatography(HPLC).The results showed that the optimal condition were 0.005 mol/L ethane sulfonic sodium—methanol—acetic acid(75∶25∶1) as mobile phase,the flow rate of 1.0 mL/min,UV detection at 280nm and column temperature at 30 ℃,and in this condition the vitamin B6,B2 and B1 were separated commendably and the contents determined more better.The relative standard deviations of detection were 0.45%~1.57%,the recoveries were 95.07%~98.41%.and the correlation coefficients for the linear equations was very high(r>0.999).The data made clear that this method have higher accuracy and veracity.By measuring vitamin B6,B2 and B1 of traditionally fermented milk samples(kurut,traditionally fermented goat milk,koumiss) from QingHai province,it was also found that the content of vitamin B6,B2 and B1 had obvious differences in different samples.
