牛膝精对辐照小鼠粒-单系造血功能的影响

【摘要】 目的探讨牛膝精(Extract of Achynanthis Bidentatae,ABE)对辐照小鼠粒-单系造血功能的影响及作用机制。方法采用造血祖细胞体外培养和体内扩散盒植入技术,观察牛膝精对辐照小鼠外周血白细胞(white blood cell,WBC)、骨髓有核细胞(bone marrow cell,BMC)及其分裂指数和骨髓粒-单系造血祖细胞(colony forming unit-granulocyte/macrophage,CFU-GM)的影响。结果辐照用药组(50mg/l、100mg/l、200mg/l)的WBC、BMC总数均较辐照对照组(0mg/l)明显提高,体内培养的CFU-GM产率也明显高于辐照对照组,而体外培养未显示刺激活性。结论牛膝精可刺激体内粒-单系造血祖细胞的分化与增植。对骨髓具有保护作用。

【Abstract】 Objective To study the Extract of Achynanthis Bidentatae(ABE) on hemopoieisis of CFU-GM in irradiated mice and its mechanism.Methods Using techniques of hemopoietic progenitors in vitro and diffusion chamber implantation invivo,the authors observed the effects of Extract of Achynanthis Bidentatae(ABE)on white blood cell(WBC)、bone marrow cell(BMC) and colony forming unit-granulocyte/macrophage(CFU-GM).Results it is shown that the number of WBC and BMC in the ABE(50mg/l、100mg/l、200mg/l)and the number of CFU-GM in diffusion chamber in vivo , were significantly higher than that of irradiated control group,but there was no stimulation activity in vitro on irradiated mice.Conclution ABE can stimulate the proliferation and differentiation of the CFU-GM and may protect bone marrow of irradiated mice.
