稻瘟病菌突变体Y34-0453的表型分析和T-DNA插入位点的定位

【摘要】 对稻瘟病菌T-DNA插入突变体Y34-0453的表型分析,发现其菌落黄化,气生菌丝减少,产孢量增加,不能侵染正常的感病水稻品种,但可以侵染具有微伤口的水稻叶片,分生孢子接种洋葱皮没有形成侵染钉。初步认为是该突变体附着胞不能正常分化,从而不能正常产生侵染钉致使致病力丧失。通过DW-ACP-PCR,获得了T-DNA插入的侧翼序列。测序并进行比对的结果表明,T-DNA插入在Ⅱ号染色体的5.184超级重叠群(Supercontig5.184)中的未知蛋白编码基因(MGG02065.5)下游1773bp处,T-DNA插入位点处可能有个远程调控元件。

【Abstract】 The phenotypes of a T-DNA insertion mutant Y34-0453 of Magnaporthe grisea were analyzed. Compared with the wild type,the mutant Y34-0453 had an etiolated colony with less aerial mycelium but more conidia. It was completely non-pathogenic to the normal sensitive host,but could infect its wounded leaf. Furthermore,it was unable to form penetration peg on the onion epidermis,which may result in loss of pathogenicity. Flanking sequences of the T-DNA insertion site on the genome DNA was amplified by the DW-ACP-PCR technique. The blast result showed that the insertion position of T-DNA was located at 1 773 bp in the downstream of a hypothetical gene (MGG 02065.5) in Supercontig 5.184,chromosome Ⅱ. There may be a long distant regulatory element in the T-DNA insertion site.
