铁皮石斛的组织培养与快速繁殖

【摘要】 以铁皮石斛(Dendrobium candidum Wallich ex Lindley)无菌苗分成的茎段、带顶芽的茎段和根蔸3类外植体在MS+6-BA0.5mg/L+NAA0.2mg/L上均能诱导出丛芽,只有根蔸能诱导原球茎,且诱导丛芽倍数高于其它两类;在此培养基上原球茎的增殖系数高于其它组合的培养基;温度对原球茎继代增殖有明显影响,(25±1)℃最适合原球茎的生长繁殖;温度对生根的影响较为明显,较低的温度(23℃)有利于减少分生苗的数量,形成健壮高大的试管苗,适量的香蕉能促进形成粗壮的根和苗。

【Abstract】 Clusters of shoots were induced from the explants of Dendrobium candidum Wall. ex Lindl: shoot section, shoot section with top bud, and roots when cultured in the medium of MS+6-BA 0.5 mg/l+NAA 0.2 mg/l. Protocombs were induced in the medium only from the explant roots, and the explant roots had higher inducing frequency of clusters than the other two explants. The reproducing rate of protocombs was much higher in this medium than any other mediums. The temperature gave an apparent impact to the subculture of the protocombs. The optimum temperature for the growth ant propagation of the protocombs was found to be (25±1)℃. The lower temperature was in favor of reducing the number of plant regeneration and establishment of healthy seedlings. Mashed banana promoted the establishment of strong roots and robust seedlings of D. candidium.
