【摘要】 目的:检测卵巢癌组织和患者血清中肽基精氨酸脱亚氨酶4(PADI4)和瓜氨酸化抗凝血酶(citrullinated antithrombin,cAT)的表达情况。方法:RT-PCR和实时荧光定量PCR检测11例卵巢癌组织PADI4mRNA表达;ELISA方法检测29例卵巢癌患者、11例卵巢癌手术后患者和23名正常人血清中PADI4和cAT表达。结果:PADI4mRNA在卵巢癌组织中的阳性表达率82%,表达量的中位数为1.57×102copies/mL;卵巢癌组(0.5053±0.0477)和手术后组(0.3941±0.0351)患者血清中PADI4表达水平高于正常对照组(0.1488±0.0148)的表达,P<0.05;卵巢癌组(0.442±0.077)和手术后组(0.3742±0.0833)患者血清中cAT表达水平高于正常对照组(0.2156±0.0116)的表达,P<0.05;卵巢癌组PADI4与cAT表达水平呈明显相关,P<0.01。结论:PADI4和cAT在卵巢癌中表达水平升高且两者密切相关,提示PADI4可能通过使抗凝血酶瓜氨酸化而参与了卵巢癌的发生发展过程。更多还原

【Abstract】 OBJECTIVE:To investigate the expressions of PADI4 and cAT in ovary carcinoma.METHODS:The expression of PADI4 transcripts in ovary carcinoma tissues(n=11) was measured by RT-PCR and Taqman real-time quantitative PCR.Expressional levels of PADI4 and cAT in sera of patients with ovary carcinoma(n=29),sera of the patients after excision surgery(n=11) and sera of the health volunteers(n=23) were measured by ELISA and Sandwich ELISA with absorbance at 405 nm.RESULTS:PADI4 mRNA was detected in 82 % of the ovary carcinoma and the mean level of the expression was at 1.57×102 copies/mL in the samples.The mean levels of PADI4 in ovary carcinoma group,after surgery group and control groups were 0.505 3±0.047 7,0.394 1±0.035 1 and 0.148 8±0.014 8,respectively.The mean levels of cAT in the 3 group were 0.442±0.077,0.374 2±0.083 3 and 0.215 6±0.011 6 in that order.Statistics analysis revealed a positive correlation between the expressional levels of PADI4 and cAT in ovary carcinoma group(P<0.01).CONCLUSION:PADI4 and cAT can increase expressions in blood of patients with ovary carcinoma,which support PADI4 may be involved in carcinogenesis and development of the tumor by citrullination of antithrombin.更多还原
