胰腺癌胰液肽表达谱变化的分析

【摘要】 目的:分析正常胰液与胰腺癌胰液多肽表达谱的差异。方法:用ZipTipC18提取胰液样品多肽,采用基质辅助激光解析离子化飞行时间/飞行时间质谱技术(MALDITOF/TOFMS)对胰液的多肽表达谱进行肽质量指纹图谱(PMF)分析,对获得的数据进行标准化处理,对高丰度的差异肽段进行TOF/TOF分析,建立胰腺癌胰液的诊断模型。结果:通过质谱分析,得到了反映胰腺癌胰液多肽组成的特征图谱,通过TOF/TOF分析,一个胰腺癌胰液相关的肽段鉴定为fibrinogenα链前体,氨基酸序列为DS-GEGDFLAEGGGVR。结论:ZipTipC18富集胰液样品后直接进行MALDI-TOF/TOFMS分析的技术,可应用于快速扫描与胰腺癌有关的多肽表达谱,建立胰腺癌胰液的多肽诊断模型,具有较好的临床应用价值。

【Abstract】 OBJECTIVE:To analyze the differential peptidomic profiles of pancreatic fluids between normal control and tumor.METHODS:Samples were handled directly by ZipTip C18 and PMF analysis was performed by MALDI TOF/TOF MS.The acquired data was standardized and peptides of interest was identified by TOF/TOF analysis.RESULTS:The peptidomic profiles which reflected the protein composition was acquired by MS analysis.By means of TOF/TOF analysis,one peptide in the pancreatic fluid from tumor was identified as fibrinogen α chain precursor.CONCLUSIONS:The method that samples was enriched by ZipTip C18 and followed by MALDI-TOF/TOF MS analysis,allows the fast screen of potential peptide/protein markers in the body fluid.The peptidomic profiles were built for the diagnosis of pancreatic cancer and its implications for diagnostics are far-reaching.
