β-NGF对人胰腺癌细胞系浸润与运动能力影响及其机制的初步研究

【摘要】 目的:探讨β-神经生长因子(beta-nerve growth factor,β-NGF)对体外人胰腺癌细胞系MIAPaCa-Ⅱ的黏附、运动及侵袭能力的影响及其机制。方法:将不同浓度β-NGF加入细胞培养液中,以过河实验和趋化运动实验检测癌细胞的运动侵袭能力,以基质浸润实验检测浸润能力,以Western Blot检测E-钙粘连蛋白(E-cadherin)和金属基质蛋白酶-2(ma-trix metalloproteinase,MMP-2)表达的变化,并与对照组比较。结果:细胞培养液中随着β-NGF浓度增加,MIAPaCa-Ⅱ细胞趋化能力明显增加,P=0.032。β-NGF组过河时间为(42.50±4.12)h,而对照组为(68.45±3.53)h,P=0.007。在基质浸润实验中,50、100和150μg/Lβ-NGF组每视野分别为(32.61±3.85)、(39.35±5.24)和(47.90±6.82),对照组每视野为(23.90±4.56),P值分别为0.008、0.004和0.002。β-NGF组中E-cadherin表达降低,MMP-2表达增加,P<0.05。结论:β-NGF可能介导了胰腺癌的侵袭转移,其机制可能是通过下调E-cadherin和上调MMP-蛋白完成的。

【Abstract】 OBJECTIVE:To investigate the roles of β-NGF on motile and invasive ability of human pancreatic cancer cell line MIA PaCa-Ⅱ,and its mechanism.METHODS:β-NGF was added to cell culture medium.Then the motile ability was tested by the crossing-river test and transwell migration assay,the invasive ability was evaluated by the matrigel invasion method,and the expressions of E-cadherin and MMP-2 were detected by Western Blot,respectively.RESULTS:The motile and invasive ability of MIA PaCa-Ⅱ obviously increased with the increase of β-NGF concentrations in cell culture medium.There were statistical differences of crossing-river time between β-NGF group(42.50±4.12) h and control group(68.45±3.53) h.By matrigel invasion test,the penetrated cells were(32.61±3.85)/field,(39.35±5.24)/field,(47.90±6.82)/field in 50,100 and 150 μg/L β-NGF groups,and(23.90±4.56)/field in control group,respectively,P<0.01.The expression of E-cadherin significantly increased,and MMP-2 decreased in β-NGF groups.CONCLUSION:β-NGF might play potentia1 roles in invasion and metastasis of pancreatic cancer,by the down regulation of E-cadherin and up regulation of MMP-23.
