依曲替酸诱导皮肤鳞状细胞癌细胞系SCL-1凋亡及其机制的初步研究
Inducement of acitretin to apoptosis of cutaneous squamous cell carcinoma cell line SCL-1 by upregulating Fas expression
【摘要】 目的:探讨依曲替酸通过上调Fas基因表达诱导皮肤鳞状细胞癌(鳞癌)细胞系SCL-1细胞凋亡的作用。方法:以体外培养的人皮肤鳞癌细胞系SCL-1为研究对象,MTT法观察依曲替酸对SCL-1细胞的生长抑制作用,Annxin V-FITC/PI法检测早期凋亡,流式细胞仪检测细胞膜表面Fas的表达情况,实时荧光定量RT-PCR方法检测Fas mRNA的表达。结果:依曲替酸能显著抑制SCL-1细胞的生长,并具有浓度及时间依赖性。浓度为1×10-5mol/L的依曲替酸能诱导凋亡,上调细胞膜表面Fas表达水平及Fas mRNA水平。结论:依曲替酸能抑制皮肤鳞癌细胞生长及诱导凋亡,上调Fas抗原表达可能是其分子机制之一。
【Abstract】 OBJECTIVE:To investigate the effects of acitretin on the apoptosis of human cutaneous squamous cell carcinoma cell line SCL-1 by upregulating Fas expression. METHODS:Human cutaneous squamous cell carcinoma cell line SCL-1 was treated with acitretin. The cell proliferation was evaluated by MTT assay, and the early apoptosis was assessed with Annexin V-FITC/PI.Cell surface protein levels of Fas were examined by flow cytometry. The real-time quantitative RT-PCR was used to detect the expression of Fas mRNA. RESULTS: Acitretin inhibited the growth of SCL-1 cells in a dose-and time-dependent manner. 1×10-5 mol/L acitretin induced apoptosis, and increased Fas expression on the cancer cell surface and mRNA level. CONCLUSION: Acitretin plays an important role in the growth inhibition and apoptosis of cutaneous squamous cell carcinoma cells,which may be related to the upregulation of Fas expression.
