细胞因子诱导的杀伤细胞体内外抗胶质瘤实验研究

【摘要】 目的:研究正常人细胞因子激活的杀伤细胞(cytokine-induced killer,CIK)对人脑胶质瘤细胞系U251的体外细胞毒活性,以及对脑胶质瘤裸鼠移植模型的体内抗肿瘤作用。方法:取正常人外周血单个核细胞(peripheral blood mononuclear cell,PBMC),通过多种细胞因子体外诱导成CIK细胞,用流式细胞仪对细胞作动态表型分析。用LDH法测定CIK细胞体外对U251的杀伤率,利用无胸腺裸小鼠U251细胞皮下移植瘤模型观察CIK细胞体内抑瘤作用。结果:CIK细胞在培养2周左右获得大量增殖,CD3+/CD56+双阳性细胞大量增殖>1000倍。体外实验证明,CIK细胞对U251有明显的细胞毒活性;体内实验表明,CIK细胞能够显著抑制Balb/c裸鼠皮下移植瘤的生长,其抑瘤率可达50%。结论:CIK细胞是一种新型和高效的免疫活性细胞,具有较强的体内外抑制胶质瘤生长的作用,有可能用于临床上脑胶质瘤的过继性免疫治疗。

【Abstract】 OBJECTIVE:To investigate the cytotoxicity of CIK(cytokine-induced killer) cells from healthy donor in vitro and their anti-tumor effects in athymic nude mice bearing U251 cells strain of glioma.METHODS:Peripheral blood mononuclear cells(PBMC) from healthy donors were induced to become CIK cells by cytokine in vitro incubation.The CIK cells were analyzed on different time points by FACS;The cytotoxicity in vitro by LDH method and the antitumor activity in vivo of the CIK were evaluated in Balb/c nude mice bearing subcutaneous U251 xenografts.RESULTS:Flow cytometry analysis showed that CD3+/CD56+ T cells expanded 1 000-fold after 2 weeks in CIK culture;CIK cells possessed high antitumor cytotoxic activity on U251 cells in vitro.In addition,CIK cells had a stronger suppressive effect on tumor growth in nude mice beating bearing subcutaneous U251 xenografts in vivo.CONCLUSIONS:CIK cells are highly efficient cytolytic effector cells which have a stronger significant suppression against growth of glioma in vitro and in vivo.The result provides an experimental basis for CIK to clinical application as a adoptive immunotherapy.
