Wnt信号通路在SD大鼠乳腺癌发生过程中动态变化意义的探讨
Dynamics of Wnt signaling pathway in the process of rats carcinogenesis of breast cancer
【摘要】 目的:观察SD大鼠乳腺癌发生过程中Wnt信号通路中各因子的动态变化情况,探讨其中与癌发生密切相关的基因。方法:利用二甲基苯蒽(DMBA)灌胃诱导SD大鼠乳腺癌的发生,在癌发生过程中动态获取SD大鼠的第4对乳腺组织标本,采用基因芯片技术对上述组织标本中Wnt信号通路相关基因的mRNA表达水平进行检测。结果:SD大鼠乳腺癌发生过程中,Wnt2、Wnt5a、Wnt9b和Tcf3、Tel4的表达出现渐进性上调,明显上调出现在诱癌后的第4~6周;APC、APC2、DDK1、DDK4和Wisp2的表达出现渐进性下调,明显下调出现在诱癌后的第6周。结论:Wnt2、Wnt5a、Wnt9b、Tcf3和Tel4表达的增加以及APC、APC2、DDK1、DDK4和Wisp2表达的减少与SD大鼠的乳腺癌发生密切相关,可能具有"开关"作用,有可能作为乳腺癌治疗或预防的新靶点。
【Abstract】 OBJECTIVE:To study the dynamics of Wnt signaling pathway in the process of rats carcinogenesis of breast cancer,and discuss the factors closed with the cancer-related genes.METHODS:Breast cancer was induced in SD rats with DMBA,in the carcinogenesis process of dynamic access,the 4th pairs of breast tissue specimens of the SD rats were collected and the Wnt signaling pathway associated gene expression levels were detected with gene chip technology.RESULTS:In the carcinogenesis process of SD rats,Wnt2,Wnt5a,Wnt9b and Tcf3,Tel4 expressions had a gradual increase,and a significant increase occurred from the 4th to 6th week after induced cancer;APC,APC2,DDK1,DDK4 Wisp2 expressions had a gradual reduction,and a significant reduction occurred in the 6th week after induced cancer.CONCLUSION:The increased expressions of Wnt2,Wnt5a,Wnt9b,Tcf3 and Tel4,and the reduced expressions of APC,APC2,DDK1,DDK4 and Wisp2 are closely related to the occurrence of breast cancer in SD rats,which may have a "switch" role and serve as the new target of treatment or prevention for breast cancer.
