骨桥蛋白及其信号通路对肿瘤侵袭调控的研究进展
Progress in research of osteopontin and its signal transduction pathway in tumor metastasis
【摘要】 骨桥蛋白(osteopontin,OPN)是一种分泌型磷酸化糖蛋白,其分子结构中含有精氨酸-甘氨酸-天冬氨酸(RGD)的受体结合序列。OPN主要通过与其受体整合素和CD44相互作用,参与肿瘤转移的多个过程。近年来,骨桥蛋白及其调控肿瘤侵袭与转移的信号通路倍受关注。OPN通过与整合素ανβ3和CD44结合作用,使多种激酶(PI3K、NIK、PLC、PKC和MAPK)磷酸化激活,从而发挥抑制凋亡和激活多种基质降解蛋白酶(uPA和MMPs)的功能,促进肿瘤细胞转移。
【Abstract】 Osteopontin(OPN)is a secreted glyco-phosphoprotein that contains a conserved Gly-Arg-Gly-Asp-Ser(GRGDS)sequence.Osteopontin mainly participates in a number of process of tumor metastasis through binding with integrin and CD44 receptors.Recently,osteopontin and its signaling pathway in the regulation of tumor invasion and metastasis have aroused concern.Osteopontin can activate the phosphorylation of several kinases(PI3K,NIK,PLC,PKC,MAPK)by interacting with integrin ανβ3 and CD44 receptors,thereby plays a crucial role in inhibiting apoptosis and activation of matrix degradation of a variety of proteases(uPA and MMPs)which ultimately promotes tumor metastasis.
