HPV感染和FHIT基因mRNA表达与非吸烟女性肺癌相关性的研究

【摘要】 目的:探讨HPV感染、FHIT基因mRNA表达与非吸烟女性肺癌的相关性,以及HPV感染对FHIT基因表达的影响。方法:采用病例对照研究方法,应用PCR技术检测HPV-DNA,相关序列,同时用RT-PCR技术检测FHIT基因mRNA缺失状况。结果:肺癌组织HPV-E6检出率高于对照组织,OR=4.379,高危型HPV感染肺组织中FHIT基因mRNA缺失率较高。结论:高危型HPV感染与女性肺癌的发生有关,HPV感染引发的肺癌可能与基因整合使FHIT基因缺失有关。

【Abstract】 OBJECTIVE: The fragile histidine triad (FHIT) gene was a frequent deletion in smoking lung cancer and HPV-associated cervical cancer. In this study, the deletion of FHIT gene mRNA in no smoking female lung cancer was inrestigated, and its relation with HPV infection was explored. METHODS: Ninety-eight lung cancer specimens and 43 normal lung tissues were examined for abnormalities of FHIT gene by RT-PCR and fragment of HPV-DNA by PCR. RESULTS: The frequencies of HPV L1, E6, and E7 DNA fragment in lung cancer were 32.65%, 20.41% and 14.29%, respectively. HPV 16 had a higher prevalence in cancer tissues than that in normal tissues (P=0.047), In all the 20 cases of HPV 16 infection, there were 17 cases in which the deletion of FHIT gene mRNA was found (OR=4.379). The deletion of FHIT gene mRNA in lung cancer was not significantly correlated with the pathological grading, lymph node metastasis, clinical stage and distant metastasis, but correlated with histology classification. CONCLUSIONS: The deletion of FHIT gene mRNA is correlated with the carcinogenesis of lung tumor. HPV infection contributes to lung cancer in the no smoking female. Deletion of FHIT gene might be involved in carcinogensis of HPV related lung cancer.
