EGF对瘢痕成纤维细胞影响的实验研究

【摘要】 目的通过探讨细胞因子EGF对瘢痕成纤维细胞生物学行为的影响,研究EGF在瘢痕形成中的作用机制,为临床预防和治疗瘢痕提供分子学水平及病理学水平的理论依据。方法采用倒置显微镜、MTT测定、流式细胞术,对瘢痕成纤维细胞及正常皮肤成纤维细胞进行体外培养,观察、分析细胞因子EGF对瘢痕成纤维细胞生物学行为的影响。结果倒置显微镜、流式细胞术以及MTT对体外培养瘢痕成纤维细胞和正常皮肤成纤维细胞的测定结果显示,与相应的对照组相比,加入细胞因子EGF后,体外培养的瘢痕成纤维细胞和正常皮肤成纤维细胞均明显增殖,且凋亡水平降低,显示EGF对成纤维细胞起着明显的促增殖作用。结论细胞因子EGF对成纤维细胞的生长趋势与增殖活性有明显的促进作用,刺激了胶原纤维的合成、增生和沉积。因此,EGF可促进创伤的早期修复,但EGF的过量应用有可能导致增生性瘢痕的形成。

【Abstract】 Objective To explore the mechanism of cytokines EGF for the scars,and to provide molecular and pathology theory for prevention and treatment of scars.Materials and Methods Fibroblasts of scars and normal skins were cultured in vitro. The results were observed and analyzed by light inverted microscope(LM),3-(4,5-dimethyl-thiazo l-2-yl)-2,5ciphenyltetrazolium bromi -de (MTT) assay,flow cytometry (FCM) to determine the effect of EGF on biological behavior of scars.Results In vitro:the results were observed by using LM,FCM and MTT assay. Compared to the control groups,proliferation of fibroblast cells increased significantly and lower level of apoptosis was observed when EGF was added to the cultures. Conclusions EGF can significantly promote the growth trends and proliferation of fibroblasts. These results demonstrate that EGF is beneficial for wound healing at early stage. However,it may lead to pathological scars formation if applied excessively.
