植物激活蛋白Ap36在苏云金芽胞杆菌的表达及抗病作用

【摘要】 将来源真菌交链孢(Alternaria spp.)的植物激活蛋白(activator protein)基因ap36克隆到pBMB982-304质粒slh(S-layerhomology)motif的下游,构建slh::fap融合基因表达载体,转化苏云金芽胞杆菌(Bacillus thuringiensis)无质粒突变株BMB171,获得成功表达。28℃培养24h重组菌发酵液处理番茄(Lycopersicum esculentum)幼苗,90min后,番茄叶片过氧化物酶(POD)和脯氨酸含量比对照分别提高了57.14%和131.59%;4d后苯丙氨酸解氨酶(PAL)含量较对照也有所提高,表明发酵液诱导番茄产生抗逆性反应。经发酵液处理的马铃薯(Solanum tuberosum)片,对马铃薯软腐病表现出很强的抗病性。

【Abstract】 The ap36 gene from Alternaria spp. was sub-cloned into the downstream of slh (S-layer homology) motif of the pBMB982-304, the expressed vector with the fusion gene slh::fap was constructed and transformed into Bacillus thuringiensis plasmid-free derivative strain BMB171. Result showed that the SLH::Ap36 fusion protein was successful expressed. Tomato (Lycopersicum esculentum) chips treated with cultured recombinant strain at 28 ℃for 24 h , showed that the activity of peroxidase and the amount of proline of tomato leaves were increased 57.14% and 131.59%, respectively, compared to control after treated for 90 min,and the activity of phenylalanine ammonia lyase was also higher than that of control after treatment for 4 d. Furthermore, the treated potato (Solanum tuberosum)chips showed higher resistance to rot disease caused by Erwinia corotovora SCG1 compared to control.
