香水月季类原球茎(PLBs)途径再生植株的研究
Regeneration of Rosa odorata via protocorm-like-bodies
【摘要】 以香水月季(Rosa odorata)的嫩叶为外植体,采用MS基本培养基,添加不同质量浓度的2,4-D、TDZ和IBA,研究了类原球茎再生植株的效果。主要步骤如下:1)嫩叶在MS+2,4-D1.0 mg/L+蔗糖30 g/L+Gelrite3 g/L培养基上暗培养4周,愈伤组织诱导率达100%,愈伤组织上的类根体(rhizoid)发生率最高,生成率可达71%。2)类根体在含TDZ的培养基(1/2 MS+TDZ 13 mg/L+蔗糖30 g/L+Gelrite 3 g/L)上,分化成类原球茎束,诱导率达71.0%,平均类球茎束为2.1。3)类原球茎在含IBA的培养基(MS+BA 2.0 mg/L+IBA 0.01 mg/L+GA 3 0.1 mg/L+蔗糖30 g/L+Gelrite 3 g/L)上,发育出小植株,萌发率为29.3%;不能萌发的类原球茎上有白色假珠芽产生。白色珠芽可进行增殖、脱分化、再分化出芽,其植株再生率为46.7%。4)再生苗的生根培养基为1/2MS+NAA 0.2 mg/L+蔗糖30 g/L+琼脂6 g/L,生根率为100%。5)小苗经驯化后移栽到灭菌草炭土∶蛭石(体积比为1∶1)基质上,成活率为95%。
【Abstract】 This study determined the probability of shoot regeneration via Protocorm-like-bodies(PLBs) in R.odorata.Using Murashige and Skoog(MS) supplemented with different concentrations 2,4-dichlorophenoxyacetic acid(2,4-D 0.1-5 mg/L),N-phenyl-N′-1,2,3-thiadiazol-5-ylurea(TDZ 7-20 mg/L) and Indole-3-butyric acid(IBA 0.01-0.3 mg/L),adventitious shoots induced from in vitro cultured leaflets of R.odorata were studied.The leaflets of R.odorata were induced for 4 weeks in the dark on the medium MS+2,4-D 1.0 mg/L +sucrose 30 g/L + Gelrite 3 g/L;the rate of callus formation was 100% and the highest induction rate of rhizoid was 71%.The number of rhizoid per explant was 3.0,and the average length of rhizoid was 0.47 cm.Using N-phenyl-N′-1,2,3-thiadiazol-5-ylurea(TDZ),rhizoids were differentiated into protocorm-like-bodies,and the highest differentiation was 71% on the medium 1/2MS+TDZ 13 mg/L + sucrose 30 g/L + Gelrite 3 g/L.The number of PLB clusters per explant was 2.1.The optimum medium that induced protocorm-like-bodies into shoot was MS+BA 2 mg/L+IBA 0.01 mg/L +GA 3 0.1mg/L + sucrose 30 g/L + Gelrite 3 g/L,and the frequency of regenerated plants was 29.3%.Clusters of e protocorm-like-bodies that failed to germinate into shoots developed into pseudobulbils.The secondary pseudobulbils can develop from the original pseudobulbils.The pseudobulbils may be proliferated and dedifferentiated on the different induction media,and then used to induce regenerated plants.The rate of regeneration was 46.7%.During induction,new pseudobulbils also occured.The appropriate medium for rooting was MS+NAA 0.2 mg/L + sucrose 30 g/L + agar 6 g/L,and the rate of rooting was 100%.After acclimation and transplantation,the survival rate was up to 95%.The suitable transplantation matrix for shoots was soddy soil∶vermiculite(1∶1).The regeneration of shoots of R.odorata may be induced by a new(leaflets-callus-rhizoid-PLB-regenerated shoots) method,that is PLBs.
