【摘要】 目的建立家族性甲状腺肿家系永生细胞系,为家族性甲状腺肿的深入研究提供永久的研究材料。方法应用EB病毒(Epstein-Barr Virus)转化技术,将家族性甲状腺肿家系成员外周血B淋巴细胞转化成永生化淋巴母细胞系。结果成功地为1个家族性甲状腺肿家系30个个体建立了永生细胞株,转化成功的细胞体积增大,形态多样,所有的细胞株冻存后复苏的成功率为100%。结论家族性甲状腺肿永生细胞株保存了原来个体完整的基因组,为在分子水平上揭示家族性甲状腺肿的发病机理提供了DNA材料。更多还原

【Abstract】 Objective To establish immortal lymphoblastoid cell bank of familial goiter is the base for further research of familial goiter.MethodsThe immortal lymphoblastoid cell lines of familial goiter were established by EBV(Epstein-Barr Virus) transforming B lymphocytes.Results30 immortal cell lines of familial goiter pedigree were obtained successfully.The results showed that cellular volume of these immortal cells was enlarged and their morphology was diversity with glitch.There was increasing development of cell aggregates of proliferate lymphocyte.All of the immortal cell lines were successfully revivification after been frozen in liquid nitrogen.ConclusionIt is important to establish immortal lymphoblastoid cell bank of familial goiter and provide long-term DNA materials for study of familial goiter in molecular level.更多还原
