【摘要】 研究了肉苁蓉(Cistanche deserticola)基因组DNA的分离提取方法.由于肉苁蓉组织内含有大量的多糖类及蛋白等物质,严重干扰DNA的抽提,在对其进行若干预处理之后,采用改进的CTAB法提取基因组DNA.根据外观、琼脂糖凝胶电泳检测、D260/D280的测定和PCR扩增表明,用改进的CTAB法提取肉苁蓉基因组DNA,所得DNA无论是纯度还是完整性都比较好,可以直接用于RAPD反应体系.更多还原

【Abstract】 The isolation of genomic DNA in Cistanche deserticola was studied.The tissue of Cistanche deserticola includes the massive multi-carbohydrates and the protein,it disturbs DNA extraction seriously.After carrying on certain pretreatments,the paper uses the modified CTAB to isolate and purify genomic DNA.According to the outward appearence,the agarose gel electrophoresis,the ratio of D260/D280 and the result of PCR indicated that with modified CTAB genomic DNA is quite good in the purity and integrity,can be used in the RAPD directly.更多还原
