神经干细胞纤维蛋白凝胶支架移植治疗阿尔茨海默病的实验研究
Study on treatment AD by transplanting neural stem cells and fibrin glue bracket
【摘要】 目的:观察神经干细胞纤维蛋白凝胶支架移植治疗阿尔茨海默病(AD)模型的效果以及神经元分化的情况。方法:体外分离培养大鼠胚脑神经干细胞,用纤维蛋白凝胶为支架移植入穹隆海马伞切断AD模型鼠海马,行为学测试后用免疫组织化学方法检测神经干细胞存活和分化情况。结果:支架组动物行为学的改善明显好于单纯干细胞移植组,支架组神经干细胞的存活和分化为神经元的情况均明显好于单纯干细胞移植组。结论:将纤维蛋白凝胶支架应用于神经干细胞的移植能促进干细胞的存活和分化。
【Abstract】 Objective: To observe the effect of transplanting neural stem cells and fibrin glue(FG) to treat AD model rats and the differentiation of neurons.Methods: We transplanted neural stem cells from embryobrain of rats in hippocampi of AD model rats transecting fimbria fornix with or without FG.30 days after transplanting,test these AD models’ behavior then inspect living and differentiation of NSCs by immuno-fluorescence.Results: Significant behavior improvement was observed in FG group and single NSCs group,but FG group is better than single NSCs group.The survival and effect of transplanting neural stem cells in FG group is superior than in single NSCs group.Conclusion: FG can improve the survival and effect of transplanting neural stem cells.
