牛磺酸对大鼠内毒素性肺损伤保护作用的研究

【摘要】 目的:探讨牛磺酸(taurine,Tau)对急性肺损伤(acute lunginjury,ALI)大鼠的保护作用及其可能机制。方法:48只大鼠随机分为3组:生理盐水组腹腔内注射生理盐水;另外2组腹腔内注射等量脂多糖(1ipopolysaccharide,LPS),并随后在第3组腹腔注射Tau。各组动物分别于腹腔注射后6、12、18 h将其处死,检测血清还原型谷胱甘肽(reduced gluathione,GSH)含量、肺组织超氧化物歧化酶(superoxide dismutase,SOD)活性及丙二醛(malonaldehyde,MDA)含量的改变,此外也观察肺组织形态学变化。结果:腹腔注射LPS使血清GSH含量及肺组织SOD活性较生理盐水组明显降低(P<0.01),而MDA含量则显著升高(P<0.01),并且大鼠肺组织出现了明显的炎症性改变;Tau处理组与LPS组比较,GSH含量和SOD活性显著升高(P<0.05),而MDA含量明显下降(P<0.05),同时肺组织的炎症改变也显著减轻。结论:Tau对ALI时的肺脏起明显的保护作用,其机制可能与MT清除过量的自由基有关。

【Abstract】 Objective:To explore in effect of taurine(Tau)on acute lung injury(ALI)and its possible mechanism in rats.Methods:48 rats were randomly divided into 3 groups:control group with intraperitoneal sodium chloride,group with intraperitoneal lipopolysaccharide(LPS),group with LPS and Tau.Rats at 6 、12 、 18 hours after intraperitoneal injection were killed and the levels of serum reduced glutathione(GSH),the levels of superoxide dismutase(SOD)and malonaldehyde(MDA)in lung tissue were detected.The histological changes in lung tissues were observed.Results:In group with LPS,the level of serum GSH and the activity of SOD in lung tissue in group with LPS was lower,but the level of MDA was higher than that in control group(P<0.01),the inflammational changes was significant.Compared with group with LPS,the level of GSH and the activity of SOD was higher(P<0.05),but the level of MDA was lower in group with LPS and Tau,which showed less severy inflammation in lung tissues.Conclusion:Tau plays a role in protecting lung from ALI,its mechanism may involve in clearing excess free radical by MT.
