应用基因芯片筛查宫颈癌的相关基因

【摘要】 目的;应用基因芯片研究子宫颈癌相关基因群的表达和功能,探讨基因芯片技术在筛查宫颈癌相关基因中的应用价值。方法:①标本的收集和处理;②总RNA的提取和质量判定;③制备探针;④芯片杂交;⑤检测与分析。结果:3例标本共同差异表达的基因共12条,表达上调3条,表达下调9条。这些基因主要涉及细胞凋亡、肿瘤转移、信号传递等多个方面。结论:①宫颈癌的发生和发展是多个基因的结构和功能异常改变的结果;②运用基因芯片技术可以快速、高通量、高敏度的筛查子宫颈癌的相关基因。基因芯片技术在宫颈癌的分子机制的研究中有着很高的应用价值。

【Abstract】 Objective:To investigate the differentially expressed gene in cervical squamous cell carcinoma use DNA microarry,explore the significance of DNA microarry in expediting our understanding of the expression pattern of cervical cancer.Methods:①Collecting and dealing with the samples.②Picking up mRNA and judging the quality of the mRNA.③Preparing hybridization probes.④Hybridization.⑤Checking and analysis.Results: By applying this chip assay to the cancerous tissues and the para-cancerous normal tissues,we identified 12 differentially expressed genes in all the 3 cancer samples,and we further identified 3 upregulated and 9 downregulated genes for functional analysis.Conclusions:①Cervical squamous cell carcinoma is the result to which balance of many genes expression were broken.②cDNA microarray is a powerful strategy to indentify novel cancer associated genes,and it can rapidly explore the roles of the novel genes in clinical cancer rapidly and efficiently.we can investigate the genes in big flux with cDNA microarray.
