不同氮源对绵羊瘤胃内固液相纤维素酶活的影响

【摘要】 本论文旨在研究不同氮源对绵羊瘤胃内固、液相纤维素酶活的影响。选择体况良好,体重50kg左右的苏尼特绵羊9只,随机分成3组,分别饲喂以鱼粉、棉粕和豆粕为主要氮源的日粮。试验分别用瘤胃内容物(液体部分)和瘤胃内容物经两层纱布过滤后的残渣(固相部分)制取酶液,采用DNS(3,5-二硝基水杨酸)还原糖法测定酶活。在39℃,pH6.8,反应1h,煮沸5min,冷却5min条件下,于530nm处比色测定瘤胃内CMC(羧甲基纤维素酶)、滤纸酶、果胶酶、葡萄糖苷酶和木聚糖酶的活性。结果表明:氮源对绵羊瘤胃固相纤维素酶活影响显著,Ⅱ组(棉粕)的滤纸酶、果胶酶、葡萄糖苷酶和木聚糖酶的活性显著高于Ⅰ组(鱼粉)(p<0.05)。固、液相部分来源的纤维素酶活差异很大,固相部分的纤维素酶活数值大概是液相部分的10倍。

【Abstract】 This paper was conducted to investigate the effects of different dietary nitrogen sources on solid and liquid fraction of cellulase in rumen of sheep.9 healthy,weighting about 50Kg sheep were randomly allocated in three groups.The three groups were daily fed on the feeds with dietary nitrogen sources of fish meal,soybean meal or cottonseed meal respectively.The enzyme was extracted from rumen content(liquid phrase)or residue resulting from filtrating rumen content through two layers cheesecloth(solid phrase).The enzyme activities were tested by the DNS reducing sugar method.After enzymatic reaction at the reaction conditions of 39℃,pH 6.8,1h reaction period,5 min of boiling and 5 min of cooling,the enzyme activities CMC,filter paper enzyme,pectic enzyme,indican enzyme and xylanase were tested by colorimetric determination at 530 nm.The results showed that nitrogen sources significantly influence enzymatic activity of solid phrase cellulase of rumen.The enzymatic activities of filter paper enzyme,pectic enzyme,indican enzyme and xylanase were significant higher for group Ⅱ than for group Ⅰ(P<0.05).In addition,the enzymatic activities were significantly different between liquid and solid phrase cellulases.The enzyme activity of solid phrase cellulase was at least ten times better than that of liquid phrase.
