与黄瓜耐高温QTL连锁的分子标记分析
Molecular Markers Linked to High Temperature Resistance QTLs in Cucumber
【摘要】 耐高温性状对黄瓜的夏季生产具有重要意义.用耐高温自交系863-7和不耐高温自交系863-6构建F2群体.在生长箱中对群体进行高温胁迫处理一个月,测定其相对生长综合值作为耐高温性状值.用62对SSR引物和132对SRAP引物组合对亲本进行多态性筛选,8对SSR引物和71对SRAP引物组合呈现出多态性.对多态性的标记进行群体分组分析和选择性基因型分析,发现1个SSR标记和9个SRAP标记与黄瓜耐高温QTL连锁,对表型的贡献率在6%~17%.Mapmaker分析发现7个标记(CSCT335、ME10EM2-350、ME11EM2-640、ME11OD3-230、DC1OD3-150、ME8EM10-590和ME8EM7-590)位于同一连锁群,2个标记(ME1EM6-670和ME9EM6-650)位于另一连锁群,标记ME9EM1-210不与其他标记连锁,它们代表的3个QTL累计的表型贡献率达32.3%.
【Abstract】 High temperature resistance is very important in summer cucumber production. Two inbred lines, 863-7 (heat-tolerant) and 863-6 (heat-susceptible), were used to construct F2 population. As a measure of heat tolerance, the relative growth (RG) was determined after treated in the growth chamber with high temperature for one month. Sixty-two SSR primer pairs and 132 SRAP primer combinations were used for selection of parental polymorphism, among them 8 SSR primer pairs and 71 SRAP primer combinations were polymorphic. Bulked segregant analysis and selective genotyping were used to screen the candidate markers. After analyzed within the F2 population, 10 markers including 1 SSR marker and 9 SRAP markers were found to be associated with high temperature resistance QTL, each accounted for 6 %~17 % of phenotypic variation. Mapmaker analysis revealed two linkage groups existed among 10 markers, one comprising 7 markers (CSCT335, ME10EM2-350, ME11EM2-640, ME11OD3-230, DC1OD3-150, ME8EM10-590 and ME8EM7-590)and another comprising two markers (ME1EM6-670 and ME9EM6-650) whereas the marker ME9EM1-210 were unlinked. These marker representing 3 QTLs accounted for 32.3 % of total phenotypic variation.
