多环境因素全面正交作用对铜绿微囊藻生长的效应研究
The overall orthogonal design study of multifactor interaction on the growth of Microcystis aeruginosa in the presence of adnascent Pseudomonas sp.
【摘要】 应用全面正交设计试验方案,研究与附生细菌共生时,温度、pH值、氮磷比三因素的不同水平组合与铜绿微囊藻生长的动态关系.结果表明,在环境温度25℃、氮磷比40∶1、初始pH为11时,铜绿微囊藻的最终生长量最大;温度25℃、氮磷比16∶1、初始pH为8.2时次之,微囊藻细胞的比增长速率相差约1.9%.统计分析结果显示,温度和初始pH值是影响铜绿微囊藻生长状况的显著因素.低温(10℃)显著抑制微囊藻的增长,低温时铜绿微囊藻对其他环境因子(pH值和氮磷比)的变化基本无响应;低pH值和较低氮磷比的组合也能明显抑制微囊藻生长,藻细胞不增长甚至处于负增长状态,此时温度对铜绿微囊藻比增长速率和最终生物量的影响程度也相应降低.
【Abstract】 Microcystis aeruginosa is a famous bloom-forming cyanobacteria,which often occurs in eutrophic lakes,such as Tai Lake.Mathematical models were established for the relations between the biomass of Microcystis aeruginosa in the presence of Pseudomonas sp.and different treatments of N/P ratio,pH and tempreture in this paper.Three levels of each factor were set.After 16 days’ combination cultivation,the results were as follows: the maximum biomass of M.aeruginosa occurred when N/P ratio was 40∶1,pH was 11 and tempreture was 25℃,and N/P ratio was 16∶1,initial pH was 8.2 and tempreture was 25℃ the next.The specific growth rates of M.aeruginosa had slight difference.Microcystis aeruginosa could maintain normal growth at temperature from 18℃ to 25℃,with the optimum temperature of 25℃.The ANOVA and the stepwise multiple regression statistical method were used to calculate the difference among three levels of the single factor,and the relation between the growth of M.aeruginosa and these environmental factors.Statistical analysis led to the results that the effect of low temperatures(10℃) had marked difference with that of middle and high temperature(18℃,25℃).What is more,tempreture and initial pH among the three factors had the notable influence on the growth of M.aeruginosa,which was characterized by the specific growth rates and final biomass.While N/P ratio had the minimal effect compared with pH and temperature.Because pH was influenced to be changing during the growth of M.aeruginosa,tempreture had the steering effects on the growth of M.aeruginosa.Low tempreture inhibited the increment of algal cells,and low N/P ratio and low pH had the similar effects.Noticeably,once there was a steering-restraining factor,other factors had slight or no effects on the growth of Microcystis aeruginosa.
