一株喹啉降解菌的分离与鉴定

【摘要】 为了研究石油污染土壤中含氮杂环化合物的降解情况,本文采用选择性富集培养的方法,以喹啉作为目标污染物,从45份石油污染土壤样品中,分离得到155株降解喹啉污染物的高效降解菌株,其中降解效率最高的—株喹啉降解菌命名为Q5,在72h内对于浓度为500mg·L-1喹啉的降解率达到72.6%。并通过形态学、生理生化试验和16SrDNA序列比对分析对Q5进行了鉴定。结果表明,株菌Q5为革兰氏阳性菌,在LB平板上菌落呈圆形,边缘整齐,乳白色,表面光滑,有黏稠感,是发酵乳酸杆菌(Lactobacillus),属酵母菌,与Lactobacillus fermentum同源性最高,为99.6%。

【Abstract】 In order to investigate the biodegradation of nitrogenous heterocyclic compounds in petroleum contaminated soil,155 strains of high-efficient quinoline degradation bacteria were isolated from 45 petroleum contaminated soil samples by selective enrichment culture when quinoline was chosen as module contaminants.The most high-efficient degradation was named Q5,its degradation rate could reach 72.6% within 72 hours when the concentration of quinoline was 500 mg·L-1.At last,Q5 was identified with 16S rDNA sequence,morphological,biochemical and physiological experiments.It was found that strain Q5 was gram-positive bacteria whose colonies were round,milky white,glossy,viscous and had regular edge on the LB plate.At the same time,the colony was Lactobacillus,a kind of yeast,sharing the highest homology(99.6%) with Lactobacillus fermentum.
