非甾体抗炎药对中波紫外线诱导的小鼠皮肤肿瘤的影响
Effects of nonsteroidal anti-inflammatory drugs on ultraviolet-B induced skin cancer
【摘要】 目的:确定口服特异性COX-2抑制剂(塞来昔布)及非特异性COX-2抑制剂(吲哚美辛)对于UVB所致小鼠背部皮肤肿瘤数目及体积的影响。方法:选用健康雌性BABL/C小鼠,应用窄谱UVB对其背部剃毛部位皮肤进行照射33周建立小鼠皮肤肿瘤模型,对口服塞来昔布及吲哚美辛与未接受NSAIDs干预治疗小鼠皮肤肿瘤发生的数目及体积进行比较。结果:与未接受NSAIDs干预治疗小鼠相比,各用药组小鼠皮肤肿瘤数目及发生率均有不同程度下降,而且差异均有统计学意义(P<0.05);塞来昔布大剂量口服有抑制肿瘤体积增长的作用。结论:口服塞来昔布、吲哚美辛均可不同程度地抑制慢性窄谱UVB照射所致小鼠皮肤肿瘤的发生。大剂量塞来昔布口服对肿瘤的体积增长有抑制作用。
【Abstract】 Objective:To determine the effect of nonsteroidal anti-inflammatory drugs (NSAIDs) on the number and size of skin cancers induced by narrow band UVB irradiation for 33 weeks. Methods: The back skin of healthy female BABL/C mice was irradiated by chronic UVB to establish a model of skin tumor. The difference in number and volume of skin tumors in the groups treated either with or without NSAIDs was compared. Results: Compared with untreated mice, the tumor numbers in all NSAIDs-treated groups were significantly reduced (P<0.05). Significant inhibition of the size of the tumors was only observed in the high dose celebrex-treated groups. Conclusion: NSAIDs such as celebrex and indomethacin can significantly inhibit UVB-induced carcinogenesis in the murine skin, but only celebrex can inhibit the sizes of the tumors.
