壮肝逐瘀煎对肝纤维化大鼠NF-кB mRNA表达的影响

【摘要】 目的:观察肝纤维化大鼠肝组织中核转录因子-kB(NF-кB)mRNA的表达及壮肝逐瘀煎的干预作用,探讨壮肝逐瘀煎在分子机制调控方面抗肝纤维化的作用机理。方法:将100只Wistar大鼠随机分为正常对照组20只、观察组80只,参照相关文献复制大鼠肝纤维化模型,造模过程中,于第4周末随机处死正常对照组及观察组大鼠各5只,证实肝纤维化形成后,将观察组其余大鼠随机分为模型组、壮肝逐瘀煎大、中、小剂量组及秋水仙碱对照组,分别予生理盐水、壮肝逐瘀煎大、中、小剂量及秋水仙碱灌胃,日1次;12周后处死大鼠,观察肝脏组织病理学变化,采用RT-PCR法检测肝组织中NF-кB mRNA的表达。结果:NF-кB mRNA在肝纤维化大鼠模型中表达增加,且壮肝逐瘀煎可以明显抑制NF-кB mRNA的表达,改善肝组织的纤维化程度。

【Abstract】 Objective:To observe the expression of nuclear factor-кB(NF-кB) in the hepatic tissue of hepatic fibrosis rats and the intervention of Zhuanggan Zhuyu Jian,and to investigate the suppressive effects of Zhuanggan Zhuyu Jian on hepatic fibrosis in rat model. Methods:Selects health clean level Wistar rat 100,takes 20 to take the normal control group and take 80 to the control group. The rats were made into models of hepatic fibrosis in accordance with some literatures. In the end of 4weeks,makes the mold rat to execute 5 to confirm stochastically HF forms. The other mold rats were divided into five groups randomly,the model group,Zhuanggan Zhuyu Jian high,middle,low dose group and colchicines group. The groups were treated with NS,Zhuanggan Zhuyu Jian high,middle,lowdose and colchicines respectively. After 12 weeks executing,to observe the rats’ histopathological change of liver,and to detect the express of NF-кB by RT-PCR method. Results:The express of NF-кB were increased in the rat model,and inhibited in the Zhuanggan Zhuyu Jian groups obviously. The Zhuanggan Zhuyu Jian can improve the degree of Hepatic Fibrosis.
