【摘要】 目的观察β-连环蛋白在转染p120ctn同工蛋白3A的肝癌细胞中的表达变化;为通过这一途径实施肿瘤的基因治疗提供实验依据。方法对BEL-7404人肝癌细胞进行稳定转染p120ctn同工蛋白3A,然后用激光共聚焦显微镜和免疫印迹技术,检测β-连环蛋白在细胞中的表达情况,同时检测细胞黏附、细胞迁移以及增殖能力的改变。结果肝癌细胞转染p120ctn同工蛋白3A后,促进了β-连环蛋白与上皮钙黏蛋白的结合,表现为其与上皮钙黏蛋白的沉淀量增加,在细胞膜上的表达增强,在细胞核中的表达趋向减弱;细胞黏附能力增强,迁移及增殖能力下降。结论转染p120ctn同工蛋白3A,能弱化β-连环蛋白在细胞核中的表达量,削弱了BEL-7404人肝癌细胞的恶性行为,是调控β-连环蛋白癌基因表达的有效途径之一。

【Abstract】 Purpose To observe the expression changes of the β-catenin in hepatoma cells transfected with p120ctn 3A and to provide experimental evidence for possible gene-therapy through this methodology. Methods The p120ctn isoform 3A was stably transfected in BEL-7404 human hepatoma cells and then the effects of β-catenin subcellular localization was investigated by immunoblotting and confocal microscopy. Cell adhesion, cell migration and proliferation were also studied. Results It is found that the p120ctn reduced to some extent the β-catenin expression at nucleus and that its membrane expression and association with E-cadherin increased. Cell adhesiveness was increased but its proliferation and mobility were decreased. Conclusions The results indicate that transfection of p120ctn isoform 3A can change the malignant features of the BEL-7404 human hepatoma cells, and it is a possibly effective channel to intervene the β-catenin oncogene expression in the nucleus.
