脑心通对动脉粥样硬化兔血管细胞黏附分子-1影响的研究
Effect of Naoxintong on VCAM-1 Level in Rabbits with Atherosclerosis
【摘要】 目的:探讨脑心通对动脉粥样硬化(atherosclerosis,AS)兔胸主动脉血管细胞黏附分子-1(vascular cell adhe-sion molecule 1,VCAM-1)的影响。方法:采用免疫组化、RT-PCR检测24只食饵性新西兰兔胸主动脉的VCAM-1蛋白和基因的表达。结果:脑心通组胸主动脉VCAM-1蛋白和基因表达均减少,RT-PCR的结果显示,单纯高脂组的VCAM-1/GAPDH光密度之比(0.97±0.21)明显高于脑心通组(0.52±0.16)(P<0.05)。结论:脑心通具有保护血管内皮,减少黏附分子表达,因此对AS的治疗及预防具有重要意义。
【Abstract】 Objective:To observe the influence of Naoxintong on vessel wall VCAM-1 level in rabbits with atherosclerosis.Methods:Twenty-four New Zealand white rabbits were randomly and equally assigned 3 groups: Naoxintong group(high-lipids plus Naoxintong 0.5g/kg/d,n=8),high-lipid group(only high-lipid diet,n=8),control group(normal diet,n=8).They were fed for 4 months,the thoracic aorta were harvested.We used immunohistochemical staining to measure the alteration of VCAM-1 proten.The VCAM-1 mRNA was analyzed by RT-PCR.Result: Naoxintong treatment significantly decreased the expression of VCAM-1 proten and mRNA.The relative value of VCAM-1 mRNA in Naoxintong group was lower than high-lipid group(0.52 0.16 Vs 0.97 0.21,P<0.05).Conclusion:Naoxintong could inhibit the expression of VCAM-1.
