口腔鳞癌中间隙连接蛋白43和E-钙粘素表达及相关性研究
Study of the correlation and expression of the Cx43 and E-Cad in the oral squamous cell carcinoma
【摘要】 目的:探讨间隙连接蛋白43(Cx43)和E-钙粘素(E-cad)在口腔鳞癌中的表达及其相关性;细胞分化诱导剂全反式维甲酸(ATRA)对人舌鳞癌细胞株(Tca8113)中Cx43和E-cad表达的影响。方法:应用免疫组织、细胞化学结合免疫印迹(western blot)技术,观察口腔鳞癌组织中及ATRA对Tca8113细胞株进行培养及分化诱导后Cx43和E-cad的变化。结果:正常口腔鳞状上皮中Cx43和E-cad主要表达于细胞膜;在鳞癌组织中,Cx43和E-cad细胞膜表达与组织的分化程度、转移之间有显著性差异(Cx43:х2=7.42、12.43,p<0.05、p<0.01;E-cad:х2=7.79、9.688,p<0.05、p<0.01)。在鳞癌组织同一标本中Cx43和E-cad表达具有正相关性和一致性(r=0.577,p<0.0005);舌鳞癌细胞经ATRA诱导后,Cx43和E-cad蛋白表达增加。结论:Cx43和E-cad在口腔鳞癌的发生和发展过程中具有协同作用;ATRA可以通过上调Cx43和E-cad的表达,实现对肿瘤生长的抑制。
【Abstract】 Objective:To investigate the expression and its relationship of the connexin 43 and E-cadherin in oral squamous cell carcinoma(OSCC);to investigate the effects of connexin 43 and E-cadherin in tongue carcinoma cell line (Tca8113) by all-trans-retinoic acid(ATRA). Method:Immunohistochemical was used to detect the expressions of Cx43 and E-cad in oral squamous carcinoma. Western blot was adopted to examine the expressions of Cx43 and E-cad in tongue carcinoma cell line (Tca8113) by all-trans-retinoic acid (ATRA). Result:There was a positive relationship and unanimity between the expressions of Cx43 and E-cad in the same sample(r=0.577,p<0.0005). After treated by ATRA,the expressions of Cx43 and E-cad were increased in Tca8113. Conclusion:The expressions of Cx43 and E-cad are coordinate with the induce and advance of OSCC. ATRA can achieve up-regulation of Cx43 and E-cad,which should be responsible for inhibited of tumor cell growth.
