反义寡核苷酸与血管内皮生长因子对血管成形术后再狭窄的影响

【摘要】 目的探讨反义寡核苷酸和血管内皮生长因子联合应用对血管成形术后再狭窄的影响。资料与方法中国大耳白兔56只,分为8组,每组7只;髂总动脉球囊拉伤后,分别给予反义寡核苷酸或/和血管内皮生长因子,取病理切片对再狭窄程度进行评价;采取免疫组织化学及Western Blotting以细胞增殖抗原的表达对反义寡核苷酸和血管内皮生长因子的效果进行评价。结果反义寡核苷酸与血管内皮生长因子各组均能减轻球囊损伤后再狭窄,反义寡核苷酸与血管内皮生长因子联合应用显著减轻再狭窄且两者间无交互作用(P>0.7)。结论反义寡核苷酸与血管内皮生长因子联合应用显著减轻再狭窄且两者之间无交互作用。

【Abstract】 Objective To investigate the effect of combined use of antisense oligonucleotide (AS-ODN) and vascular endothelial growth factor (VEGF) on restenosis after balloon angioplasty. Methods Fifty-six Chinese rabbits were divided randomly into eight groups with 7 rabbits in each group. AS-ODN, VEGF or combined use of both AS-ODN and VEGF was administrated by local delivery immediately after balloon angioplasty. The degree of restenosis after balloon angioplasty was evaluated on histologic sections of the target artery. Immunohistochemistry techniques and Western Blooting methods for the express of proliferating cell nuclear antigen (PCNA) were employed to assess the effect of AS-ODN or/and VEGF on restenosis of the artery. Results AS-ODN or VEGF could inhibit restenosis of the artery. However, combined use of both AS-ODN and VEGF could inhibit restenosis more effectively than only AS-ODN or VEGF without interaction with each other for both drugs. Conclusions Combined use of both AS-ODN and VEGF could prevent restenosis of the artery after balloon angioplasty without interaction with each other for both drugs.
