【摘要】 目的比较应用单纯直接贴壁法和密度梯度离心结合直接贴壁法两种方法分离、纯化、扩增猪骨髓间充质干细胞(MSCs)的培养效率,为临床应用提供理论依据和实验方法.方法取滇南小型猪的骨髓液用单纯直接贴壁法和密度梯度离心结合贴壁培养法两种方法分离猪骨髓间充质干细胞,体外培养扩增,绘制其生长曲线,通过显微镜观察培养细胞的生物学形状.结果直接贴壁法收集到的细胞扩增速度较慢,密度梯度离心法收集到的细胞扩增速度较快,细胞培养后成梭形.结论密度梯度离心结合贴壁培养法较单纯的直接贴壁法培养骨髓间充质干细胞的培养效率更高.体外培养的猪骨髓间充质干细胞生长稳定,增殖力较强.更多还原

【Abstract】 Objective To provide a theoretical basis for clinical and experimental methods by comparing the cultivation efficiency of the two methods that are direct adherent and density gradient centrifugation.The two methods are to separate,purify and amplify swine bone arrow-derived mesenchymal stem cells (MSCs).Methods Swine bone marrow were taken from the southern area of Yunnan province and the two methods combined were applied to culture swine bone marrow mesenchymal stem cells and proliferate in vitro,then the shapes of cell were observed through the microscope and its growth curve was drawn.Results The collected cells proliferated slowly by direct adherent,and the speed of that is faster by density gradient centrifugation,and later all of these cells exhibited spindle in shapes.Conclusion The speed of culture MSCs by density gradient centrifugation combined with adherent is faster than that of by a simple direct adherent.The MSCs which cultured in vitro grows steadily,and the proliferation is strong.更多还原
