干扰素-γ对小鼠结核病的实验研究
Experimental Studies of Effect of Interferon-gamma on Model of Mice Infected Mycobacterium Tuberculosis
【摘要】 目的:研究干扰素-γ(IFN-γ)对结核分支杆菌感染小鼠的疗效与机制。方法:将BALB/c小鼠制成结核分枝杆菌感染模型,随机分为4组,分别以IFN-γ组,左氧氟沙星组(LVFX),联合组及磷酸盐缓冲液组(PBS,对照组)治疗。检测重要脏器菌落数,观察小鼠肺组织病理改变和平均生存时间。结果:各治疗组肝、脾、肺组织菌落数均显著低于对照组,肺部损伤亦较轻,尤其联合组效果更为明显。各治疗组小鼠平均生存期均显著高于对照组(P<0.01)。结论:外源性IFN-γ可减少肝、脾、肺组织中结核分枝杆菌菌落数,减轻肺部病变,延长动物生存时间,促进Th1细胞的免疫应答,在抗结核分枝杆菌感染中起着重要作用。
【Abstract】 Objective:To study the effect and mechanism of IFN-γon resistance of BALB/c mice to Mycobacterium Tuberculosis infection.Methods:The BALB/c mice infected with Mycobacterium tuberculosis were randomly divided into four groups and were treated with IFN-γ,LVFX,IFN-γ+LVFX and PBS,respectively.The colonies of bacteria in major organs were counted,and the pathological changes in the lungs and the survival time of the mice were observed.Results:In three treated groups the bacterial colonies in the livers,spleens and lungs were markedly reduced than those in control group.And so did in the lung lesions,especially in IFN-γ+LVFX combination treated group.In the treated groups the mean survival time of the mice was longer than that of the control group(P<0.01).Conclusion:This study shows that exogenous IFN-γcould decrease the bacterial colonies in the liver,spleen and lung,and relieve the lung lesions and extend the survival time of mice infected with Mycobacterium Tuberculosis,It can also promote immune response of Th, cells.It suggested that IFN-γcould play an important role in anti-tuberculosis protection in mice.
