TTRAP低调表达方法的探讨
Research on Down-Regulated TTRAP Expression
【摘要】 目的:建立特异性低调TTRAP表达的方法,以探讨TTRAP蛋白在TNF信号通路中的重要作用。方法:设计3条针对TTRAP的siRNA,在HEK293细胞系中使用瞬时转染和蛋白印记方法进行筛选。结果:在三条针对TTRAP的siRNA中,其中有一条siRNA在55·19%转染效率下可以特异性抑制约50%的TTRAP蛋白表达。结论:提供了一种可靠的特异性低调TTRAP表达的方法,可望有助于了解TTRAP在TNF信号通路中的功能研究。
【Abstract】 Objective:To explore the effect of the TTRAP protein in TNF transduction pathway,the special method of down-regulated TTRAP expression was applied. Methods:Three putative TTRAP specific duplex siRNAs were screened in HEK293 cell line with siRNAs instantaneous infection and Western blot. Results:One of the duplex siRNA down-regulated about 50% TTRAP expression in HEK293 cell line by 55.19% transfection rate.Conclusions:This research provided a reliable method for down-regulate TTRAP expression that may improve the work on exploring the TTRAP function in the TNF transduction pathway.
