栽培稻与Oryza officinalis相似群野生稻基因组VDAC基因的比较
Comparison of VDAC Genes between Cultivated and Oryza officinalis Group Wild Rice in Genome DNA
【摘要】 根据水稻基因组文库日本晴的基因组VDAC基因的序列,设计引物分别扩增6种不同基因组的野生稻(BB、CC、BBCC及CCDD)和2种栽培稻(AA)的基因组DNA的VDAC基因片段。所有分别代表8个VDAC基因的8对引物中,除引物VDAC3外,其它7对引物均能扩增出预期大小的特异条带,其中一些片段是所有试验材料共有的,而另一些则具有明显的基因组或种的特异性。AA、BB、CC基因组均具有VDAC1、VDAC4、VDAC7和VDAC8引物的扩增位点,而DD基因组则不能确定。VDAC6的引物位点是AA基因组所特有。VDAC2引物的扩增位点存在于基因组AA、BB、DD,而CC基因组中则无此位点。而VDAC5则可区分同为CCDD的宽叶野生稻和高秆野生稻。栽培稻种与野生稻种基因组相比能扩增出更多的VDAC基因片段的试验结果,为进一步研究稻属中VDAC基因的起源及进化关系提供了基础。
【Abstract】 According to the sequence from the rice date base,8 primer pairs for 8 VDAC genes were designed.The fragments of 8 VDAC genes were amplified with the template DNA of different wild rice(with BB,CC,BBCC and CCDD genome) and cultivated rice(AA genome) through the VDAC 1-8 primers.Within the 8 primers,there were 7 primer pairs that special bands were amplified.The results showed that some VDAC fragments appeared in all studied materials,and some represent the difference between the different genomes or species.AA,BB,CC genome all have the primer sites of VDAC1、VDAC4、VDAC7 and VDAC8,and DD genome was unconfirmed.The primer site of VDAC6 was special for AA genome.Only in CC genome there was no the primer site of VDAC2.And O.latifolia and O.alta,which both with CCDD genome,could be distinguished by using the primer VDAC5.The results of the cultivated rice have more VDAC sites than the O.officinalis group wild rice supplied some basic for studying the origin and the evolution of VDAC genes in Oryza.
