替米沙坦对2型糖尿病大鼠胰腺β细胞的影响

【摘要】 目的观察替米沙坦(Tel)对高脂高果糖饲料(HFFD)喂养诱导的糖尿病SD大鼠胰岛结构和功能的影响,探讨其对糖尿病大鼠胰腺β细胞的作用。方法正常SD大鼠随机分为3组:标准饲料组(SCD)、HFFD组和HFFD+Tel组,每2周末测定空腹血糖(FSG)和空腹血胰岛素(FSI),第12周末测量体重,腹腔注射葡萄糖耐量试验(IGTT),处死大鼠,取胰腺组织称重,部分组织HE染色、免疫组织化学观察胰腺结构,另一部分胰腺组织RT-PCR检测胰腺组织中葡萄糖转运体2(Glut 2)mRNA的表达。结果HFFD组与SCD组比较,FSG和FSI水平升高,葡萄糖耐量受损,胰岛素分泌第一时相缺失,β细胞数目减少,胰腺组织Glut 2 mRNA表达降低。HFFD+Tel组与HFFD组比较,FSG和FSI显著降低,葡萄糖清除率得到明显改善,胰岛素分泌第一时相存在,β细胞数目增加,胰腺组织Glut 2 mRNA表达升高。结论Tel对糖尿病大鼠具有降低FSG和FSI、减轻胰岛素抵抗、保护胰腺β细胞的作用。

【Abstract】 Objective To investigate whether telmisartan has a protective effect on structure and function of pancreas of type 2 diabetic rats induced by high fat and fructose diet. Methods The normal Sprague-Dawley(SD) rats were randomly divided into three groups: standard chow diet(SCD) group,high fat and fructose diet(HFFD) group,and high fat and fructose diet with telmisartan(HFFD+Tel) group.Fasting serum glucose(FSG) and fasting serum insulin(FSI) were measured every two weeks.At the end of twelfth week body weight of SD rats were measured,intraperitoneal injection glucose tolerance test(IGTT) was performed,SD rats were sacrificed,Pancreatic tissue was isolated and weighed.HE staining and immunohistochemistry method was used to observe the structure of pancreas,reverse transcription polymerase chain reaction(RT-PCR) was performed to test the level of glucose transporter 2(Glut 2) mRNA in pancreatic tissue. Results Compared with SCD group,the level of FSG and FSI of HFFD group heightened,glucose tolerance impaired,the first phase of insulin secreting disappeared,the number of β cells reduced,the level of Glut 2 mRNA degraded.Compared with HFFD group,the level of FSG and FSI of HFFD+Tel group descented significantly,glucose clearance improved,the first phase of insulin secreting existed,the number of β cells increased,the level of Glut 2 mRNA augmented. Conclusions Telmisartan can lower FSG and FSI,ameliorate insulin resistance,protect the function of pancreatic β cells of type 2 diabetic rats.
